The project was focused on membrane protein from Streptomyces coelicolor – CpkF, which we believe is
responsible for active transport of a new polyketide. In my thesis I clearly proved that this protein is necessary for active efflux of the polyketide. Using the PCR targeting method I constructed S. coelicolor cpkF deletion mutants, and then complemented them by introducing cpkF gene in another site on the S. coelicolor chromosome. I constructed overproduction mutant by introducing additional copy of the cpkF gene on an integration vector to S. coelicolor wild strain. I also prepared constructs for heterologus expression of CpkF protein in E. coli and scanned cultivation conditions for protein isolation.

The aim of this project was to overexpress Neuroglobin and Cytoglobin in E. coli using pET vector system to get appropriate volume of proteins which will be useful for investigation their function, structure and interactions. And because we can not avoid of expression any other contaminent protein in bacteria I had to purified my globins using standard protein purification steps.
