  • To study the effect of active extracts of medicinal plant CBT-023 for cell cycle analysis followed by apoptosis in A549 and Differentiation therapy in  HL-60 cell line in-vitro.

(Abstract): Selection of Medicinal plant followed by sequential extraction with solvents ranging from non polar to polar was done and extracts were resolved using Thin layer chromatography to check its major components. Then, Anti-cancer activity of methanol and dichloromethane extract of medicinal plant was tested in HL-60 and A549 cells invitro. Two hallmarks of cancer therapy viz differentiation and cell cycle arrest were considered as the best way to eliminate cancer cells. Cell viability and proliferation was checked by MTT assay and 3[H] thymidine incorporation assay respectively. Differentiation in HL-60 Cell lines was assayed by Geimsa staining. Cell cycle arrest in HL60 and A549 was determined by flow cytometry and confirmed by analyzing the expression of CDK 2, 4, 6, Cyclin E, D1, D3 and cell cycle inhibitor P16 and P27 by Western Blotting. Both differentiation and cell cycle arrest leads to Apoptosis which was analysed by DNA fragmentation and confirmed by protein expression of PARP and gene level expression of Caspase 3and 8 using RT PCR.
