Ltd., Dewas.\r\nProjects Undertaken \r\n A project on Studies and Monitoring of Ambroxol and Ethylene Glycol in Benincasa Hispida Fruit Extract and Pyridoxine for Lithogenic and Antilithiatic Activity in Rats.\r\n A project on Time and Motion Study in Vial Line. \r\n A project on Nanotechnology in Modern Drug Delivery System.\r\n A project on Validation Protocol and Report. \r\n \r\nProject details\r\n\r\nStudies and Monitoring of Ambroxol and Ethylene Glycol in Benincasa Hispida Fruit Extract and Pyridoxine for Lithogenic and Antilithiatic Activity in Rats\r\n\r\nThe purpose of this study was to determine the lithogenic property of ambroxol and antilithiatic activity of Benincasa hispida fruit extract by examining the urine (for pH, calcium, sodium, uric acid, urea and oxalate) and serum (for calcium, sodium, uric acid and creatinine). Ambroxol influence on the pH level was measured in 6 rats treated with 60mg/kg (dissolved in injection water, sc., daily) during 2 weeks period. Urine was collected once in every 24 hrs and measured with a digital pH meter. The influence of Benincasa hispida fruit extract on the urine and serum biochemistry and determined by urinalysis and serum analysis. The blood was collected from the animal after euthanasia by puncturing the left ventricle.\r\n\r\nThe dynamics of pH change was measured in 4 rats treated with 60 mg/kg/24h of ambroxol. Control received 1ml of injection water sc. The urine pH decrease up to 6.24 ± 0.18 vs. 7.08 ± 0.07 mg/dl in the control (P < 0.001). The serum uric acid level increased up to 4.05 ± 0.42 vs. 2.76 ± 0.29 mg/dl (P < 0.05). The serum level of creatinine also increased to 1.22 ± 0.21 vs. 0.60 ± 0.08 mg/dl (P < 0.001). The level of oxalate in urine increased to 3.17 ± 0.41 vs. 0.72 ± 0.05 mg/dl (P < 0.001) in the group treated with ambroxol.\r\n\r\nAmbroxol withdrawal resulted in sequential urine pH decrease. 11 days after interruption of ambroxol therapy pH nearly reached the starting value.\r\n\r\nAll the values are represented with means ± SEM. The student t-test was used to compare the means, P < 0.05 was considered as significant. Comparisons are made between different parameters using one – way ANOVA to identify any differences.\r\n\r\nIn conclusion the dose of 300 mg/kg of aqueous and methanolic extract of Benincasa hispida was found effective in the treatment of nephroliths.
