PROJECTS- CENTRAL DRUG RESEARCH INSTITUTE (DIV.OF TOXICOLOGY)

 PROJECT TOPIC-Assesment of DNA damage on peripheral blood leucocytes and Macrophages after Lipopolysaccharide exposure in In vivo and In vitro systems.

Summary-Lipopolysaccharide (LPS) is an prototypic endotoxin and well known inflammation causing agent.It is regularly used by researchers to explore the pathogenesis of infalmatory diseases.LPS acts via bindind to several receptors like CD-14,TLR4/M2 and could cause the release of several proinflamatory cytokines could led the cells towards death via apoptosis.
Leucocytes are considered as the phagocytic cells of body and first affected in any pathogenesis.In the present study we explore the LPS mediated effect on peripheral blood leucocytes and macrophages via COMET ASSAY.Both In vitro and In vivo studies were performed for investigation.We also explore the effect of compound 2-oxo-pyridine on LPS mediated DNA damage,by COMET ASSAY,SINGLE CELL GEL ELECTROPHORESIS AND DNA FRAGMENTATION.

 Seminar presented on regulation of CELL CYCLE in Eukaryotic models at Molecular level in M.Sc. part I at R.G.B.C.RTMNU ,Nagpur

 Seminar presented on Recombinant DNA Technology in M.Sc.Part II at RGBC,RTMNU,Nagpur.

 Participated in “International Conference on Innovations in Biotechnology and their Applications, GLOBAL SUSTAINABLE- 2nd BIOTECH CONGRESS (2006) as volunteer and member of registration committee from 18th-21st Dec 2006.


PRACTICAL SKILLS:
 Cell Culture techniques:
Media preparation for in vitro cultures ,Trypsinization of cell lines ,subculturing ,passaging, Isolation of cell clones.

 Spectroscopic Techniques:
UV Spectroscopy, infrared spectroscopy, flourescence spectroscopy, calorimetry.

 Molecular Biology Techniques:
Standard and real time PCR, plasmid based cloning, cloning into bacterial expression and phage vectors, RFLP, isolation of genomic DNA from bacterial, animal and plant cells, isolation of plant and yeast mRNA, isolation of plasmid DNA, restriction digestion and ligation (recombinant DNA technology), transformation (heat shock) in E.coli, southern blotting, protein isolation (SDS PAGE), purification, estimation.

 Toxicological Assays:
1. Comet Assay for cell viability and cytotoxicity.
2. Hoescht assay to evaluate the apoptosis in cell lines or in vivo models.
3. DNA Fragmentation of Drug Treated cell at different concentrations.



 Neurobiological Techniques:

Short term brain culture, detection of neuropeptides using bright field and flourescence microscopy, stereotaxic implantation of the cannula and intra-cerebroventricular administration in rats, application of behavioral models with specific reference to anxiety and stress.

 Other Techniques: Animal house maintenance and animal care (Sprague dawley rats and Charles foester rats).

 Gel Electrophoresis.
 Spectrophotometer (UV – Vis.).
 Centrifuges (including cooling centrifuge)
 Plant Growth Chamber/ Tissue Culture Instruments
 ELISA Reader, CO2 Incubator, Thermal Cycler (PCR), Electrophoresis Units.



COMPUTER SKILLS : Windows XP, Vista and Linux (OS). Adobe Photoshop,
Corel Draw, Graph pad and Origin 50.
